 MY Stories

Cleistanthin

Driven by a profound interest in research, my journey began in 1992 when I joined Madurai Kamaraj University's Cancer Biology lab under the guidance of Dr. G. Shanmugam. Assigned to work on P53-like anti-oncogene and its mutations in cell culture, my childhood fascination with Indian home remedies, folk medicine, Ayurveda, and Siddha continued to influence my perspective. In January 1993, during a visit to the Palani temple with Sendurai Mani, I marveled at the unique composition of Lord Muruga's statue, made from a mixture of nine different toxic plants known as Navabhashanam.

Expressing my desire to screen such plants, I sought Mani's help in acquiring them from his village. Emphasizing the need for plants with a molecular mode of action, we identified cleistanthus collinus, a slow-killing plant, in February 1994. Conducting an early morning experiment with a methanolic extract added to Hela cells, we observed peculiar behavior in the cells. Despite facing criticism from lab seniors for seemingly unconventional work, I persisted with the intention of showcasing the results to Dr. Shanmugam.

To my surprise, Dr. Shanmugam remained calm during the observation, prompting an unexpected call in the evening. Instead of termination, he showed keen interest, asking detailed questions about the idea and the experimental process. This led to the inclusion of the work as part of my Ph.D., offering me the opportunity to explore various techniques in chemistry, biochemistry, and molecular biology across different institutes.

Over the course of my research, I performed diverse experiments, including purifying compounds with over 6000 TLC plates, purifying in large solvent separation columns, conducting spectroscopic analyses, blood biochemistry on mice receiving cleistanthin, and working on more than 38 cancer cell lines in Pune NTCC. My efforts extended to FACS flow cytometry at CCMB, Comet assay, oxidative damage assay, TBAS oxidative damage, antioxidative effects, chromosomal karyotyping, and research on mouse-induced cancer and its cure.

Notably, cleistanthin exhibited superior efficacy compared to known chemotherapy agents like Tamoxifen and Etoposide, impressing a Boston lab with its high anticancer effect and low side effects on blood cells. Despite encountering patent issues leading to project discontinuation, in the year 2000, Sendurai Mani carried the work forward by collaborating with Sathgen on cleistanthin derivatives. In 2023, the Indian government approved human trials, raising the prospect of testing on cancer stem cells.

Importance of Ph.D work in Cancer : This work was the first to show the effect of Clesitanthin compounds purified from cleistanthus collinus on cancer cells with least effect on normal cells in comparison to other leading cancer drugs. Cleistanthus collinus is a toxic plant , but purification results in removing the toxic effects and showing anticancerous activity.

This work is still motivating other researchers to work on these compounds.  https://www.alliedacademies.org/conference-abstracts-files/molecules-targeting-cancer-stem-cells.pdf  Godvari Refinieries ,  Sathgen,  a company has studied the effect on clesitanthin and its derivatives.

AMMAGENOMICS

In 2021, after visiting our parents in Chennai, we decided to return to Arizona, where we had previously registered AMMAGENOMICS. However, the COVID-19 pandemic made it challenging to return to the USA. During this time, my classmate Mr.Ganesan asked me why I wasn't contributing to the creation of COVID testing kits. For research purpose my wife Priya was instrumental is setting a lab in Chennai, and due to excellent customer service we had to employ more than 30 people and helped the community in giving 3 hours results. After this these customers turned towards us for quick blood and other diagnostic services.

OORJV

I first considered the concept of Oorj while I was in the USA over a decade ago. My journey toward healthier food choices began back in 1997 when I started consuming organic milk, as that was the primary option available at the time in Buffalo, USA. However, it wasn't until around 2000 that I had access to a more extensive range of organic foods. At that time, the terms "organic" and "non-GMO" were commonly used, but it's important to note that not all organic foods were necessarily non-GMO. If a food item had undergone genetic modification, it was considered GMO, while non-GMO or traditional heirloom referred to those that hadn't.
It's also crucial to distinguish between GMO and hybrid foods. The misconception that organic food is entirely pesticide-free is incorrect; there are specific regulations that govern which pesticides can be used in organic farming.
During the process of obtaining organic certification, questions are answered, and recommendations are made. Soil samples are rarely tested. These tests aren't typically used to certify products.
I developed several novel technologies in a USA company, not just for widely produced crops like soybeans and maize but also for processed foods. This eventually led to the non-GMO initiative. This initiative involved certifying non-GMO food through genetic analysis.
Around 2010, the market started overflowing with organic products as the awareness of organic food's health benefits grew. People began to switch to organic options. I remember a conversation with my wife in a store, where we realized that earlier, organic choices were limited to specific products like olive oil or sesame oil. However, with time, organic options became more diverse, and the question arose, "With so many options available now, how do consumers decide?"

Shop Awareness - Kapila Nalam
This led to the idea of shop awareness. I initiated the process to register in India with my friends including Thanigai, Muthu, Ripunjaya, Viola, Jayaram, Vijay, Pongalur Gomathi Ganesan (Who later took it forward as www.kapilanalam.com) .

I was particularly interested in the polyphenol content of foods, as I believed in their significant health benefits. Polyphenols are known for their anti-inflammatory and antioxidant properties, and they can delay the onset of various aging-related disorders and diseases. I started testing polyphenols, and my interest in their health benefits expanded.
In 2021, after visiting our parents in Chennai, we decided to return to Arizona, where we had previously registered AMMAGENOMICS. However, the COVID-19 pandemic made it challenging to return to the USA. After this time, continued testing foods rich in polyphenols, which eventually led to the formation of AMMAGENOMICS and OORJV. Our journey started with testing for polyphenols, and over time, we began to test a wider range of foods.
As we received more food samples from customers, we observed that adulteration was becoming more prevalent in India than in other places. This led to my decision to move to India, as I consider it vital to consume foods rich in polyphenols. We noticed that young members of our lab staff in India were consuming a lot of junk food. To encourage them to adopt healthier eating habits, I introduced OORJ-certified foods. We even conducted tests to measure polyphenol levels in their blood, which confirmed the health benefits of consuming polyphenol-rich foods.
OORJ certification ensures that the food you consume is of high quality. Unlike organic products, OORJV-certified items undergo rigorous testing, including checks for pesticide residues and adulteration. The certification aims to provide consumers with a trusted way to ensure the quality of the foods they eat.
We have identified farmers throughout India who use traditional pesticide-free methods and tested not only for polyphenol levels but also other aspects of food quality. Our goal is to provide consumers with confidence in the safety and health benefits of the food they consume.
This initiative is essential not only for organic foods but also for foods with medicinal properties, as we recognize the importance of healthy eating. We aim to make OORJ a valuable player in the global market.
AMMAGENOMICS differs slightly from OORJV and has been validated and tested by OORJV. While all the testing and examinations are performed by AMMAGENOMICS, OORJ ensures that the testing and other requirements are met, which is akin to a certification process. The difference lies in the fact that the University awards the certification.
This is how we distinguish ourselves from other testing companies, and we have plans to expand our services to the USA and other countries. The market is currently inundated with various claims about food quality, and our goal is to provide consumers with reliable information and certification. As a result, more sellers are starting to market OORJ-certified products, and our impact on the food industry is growing.
Our commitment extends beyond polyphenols to other aspects of food quality and safety. We want consumers to be certain about the foods they consume, and our rigorous testing and certification process aims to achieve this.
